Differences in Blocks

If i bought long tube headers for my stock 01 Z06 motor, if i upgraded to a 383ci, a 402ci or even a 427ci will the same long tubes hook up or will i need a custom set up?

They will fit fine. We assume they are 1 3/4" primaries.

However a larger cube engine...say a 427 works better with 1 7/8" primaries. You may want to consider going to the larger primary headers with more cubic inches of displacement....especially if the heads will be ported.
